For the sake of consistency with Table 1, alleles read from the forward DNA strand, relative to the revised Cambridge Reference Sequence (rCRS) [23, 24], unlike Figures 2 and 3 that show the state according to the actual strand targeted by the SBE assay. These results are identical to the cloned singleplex results shown in Table 1, demonstrating consistent genotyping of the Iceman by different methods. As all these results were obtained in a single multiplex reaction, credibility is given to those reactions returning the rCRS. Such results in a singleplex scenario could be explained by contamination of an individual assay by any non hg K mtDNA source.
